Development of Novel Method for Assessment of Antitumor Effect of 5-Fluorouracil, Methotrexate and Retinoic Acid on Murine Fibrosarcoma Implanted in Mouse


Abstract
The experiments were carried out to elucidate the effects of 5-fluorouracil, methotrexate and retinoic acid on marine fibrosarcoma implanted in ICR mouse.
To elucidate the relationship between anticancer drugs and antitumor effects, the feature of s phase cells was studied by flow cytometry, the metabalic status of implanted cells, PDE/ATP, PCr/Pi and ATP/Pi were studied by nuclear magnetic resonance, and the enzymatic activity of thymidylate synthetase and dihydrofolate reductase were measured with spectrophotometer.
The results were as follows ;
1. S phase fraction of tumor cells was significantly decreased in the implanted marine fibrosarcoma cells treated with 5-fluorouracil, methotrexate and retinoic acid, and synergistic effects of anticancer compounds were shown in the fibrosacoma cells treated with 5-fluorouracil and methotrexate, and 5-fluorouracil and retinoic acid simultaneously.
2. The inhibition of thymidylate synthetase activity was shown in the fibrosarcoma cells treated with 5-fluorouracil, 5-fluorouracil and methotrexate, and 5-fluorouracil and retionic acid simultaneously, dihydrofolate reductase activity was inhibited in the fibrosarc.oma cells treated with methotrexate, and 5-fluorouracil and methotrexate simultaneously.
3. The ratios of PCr/Pi and ATP/Pi were significantly increased in the fibrosarcoma cells treated with 5-fluorouracil, methotrexate and retinoie acid, and the ratio of PDE/Pi was significantly decreased in those treated with 5-flunrouracil Land methotrexate, 5-fluorourac.il and retinoic acid simultaneously.
Results obtained by ¡ÝP NMR, flow cytometry, and measurement of enzymatic activity may be valuable for evaluating antitumor effects of anticancer drugs.
